PURPOSE: Describe how to collect, transport and prep a swabbed specimen for CT/NG testing.

Specimen collection and transport:

Step 1

Endocervical and urethral swab specimens can be collected and transported in 1-3 ml transport culture media (2Sp culture transport medium, Bartels ChlamTrans, Chlamydial transport medium, SPG culture transport medium or M4 culture transport medium).

Step 2

Use only Dacron, rayon, or calcium alginate tipped collection swabs with a plastic or non-aluminum wire shaft. Do not use collection swabs with wooden or aluminum shafts for they will interfere with PCR amplification by causing inhibition.

Step 3

After sampling the cervix or urethra place swab in transport medium and seal with cap. Label specimen with patients name, test(s) requested and doctor who is ordering the test.

Step 4

Refrigerate swab if specimen transport is to be delayed for more than one hour from time of collection. Swab specimens may be transported to the test site at 18-25 degrees Celcius provided that the total time of storage and transport at 18-25 degrees Celcius is less than 1 hour.

Step 5

Swab specimens that require shipment to off-site laboratories should be shipped at refrigerated temperature as soon as possible after collection. Transport of specimens must comply with country, federal, state and local regulations for the transport of etiologic agents.

Step 6

Store swab specimens that are not tested upon receipt at the laboratory at 2-8 degrees Celcius and process within 7 days. Swab specimens that cannot be processed within 7days of collection must be stored at -20 degrees Celcius or colder and tested within 30 days of collection.
